There are several differences
Number of clients - Express -10
Extreme - 50
Number of ethernet ports - Express 1
Extreme - 3
Express can be used as an audios device for Air Tunes. Extreme cannot.
Extreme can do the following that the express cannot - Dual frequency network.
Allow sharing of disk drives.

Airport express is built small, only including one Ethernet slot in, a usb slot, and a line out slot. The Airport Extreme has a Ethernet in slot, three Ethernet out slots, a usb slot and a line out slot. If you just want to add onto your network, or add speakers, the airport express will do. If you want it to use Ethernet wires to hook up computers, or share a hard disk and/or a printer, I would recommend the airport extreme.
Hope this helps!

There IS no line out port on the Airport Extreme Base Station. Check the specs and images on the Apple Store site. If you are going to stream music via WIFI you'll need the Airport Express.
