Can I attach my new 27" Apple monitor to my 13" Macbook Pro without turning off the power first on the laptop.

Yes. Monitor connections are hot swappable. When you dis/connect a monitor, OSX will blank out the screens while it automatically adjusts to the new configuration.
Since this monitor also attaches to the MB/MBP with USB and Power, those are also hot-swappable. Obviously your laptop will run off battery power until you connect it to another power source. Also, if you have other devices attached to the monitor's USB ports, like external hard drives, they should be unmounted from the OSX desktop on the laptop first.
