Could I execute the Time Machine back up on the USB storage which connected on the AirPort Extreme Base Station thru wireless or wire?

Yes. I have a 1TB drive used for Time Machine backup, and a printer, connected to a USB hub, then to the AirPort Extreme USB port. Both printer and backup drive perform flawlessly.

Yes, you can backup both through wireless and through wire.

This is NOT OFFICALLY SUPPORTED by Apple, but that is pretty well hidden.
It works well for some, not so reliably for some, not at all for others.
This forum won't let me post a link to the Apple Technical article, so here's how to find it:
Go to apple.com (slash) support, click Mac, then Mac OSX v10.6 Snow Leopard, then select Time Machine, then Other Time Machine articles. The article in question should appear on the right, titled "Time Machine doesn't back up to AirPort Extreme Airport Disks.
If you have a problem with this configuration and call AppleCare, you'll get very little help, if any.

yes you can
1/ first step is to format the hard drive connected directly to the mac (mac os extended journaled) using the disk utility
2/then you disconnect it and connected to the base station
3/ select the hard drive in the preferences from time machine
4/ sit back and wait since it is over a wifi it is going to take a while
