Can the AirPort Extreme with a connected hard drive act as an iTunes server for multiple computers?

It can at a file server. So, on each computer - when you start iTunes, choose to start with option and select a library - and choose the file from the common shared folder.
Notice, this is different from a shared iTunes library using iTunes. A shared iTunes library needs a server serving iTunes.
Best option is to put your media file in TimeCapsule, Run any one iTunes and make it use the shared file from TimeCapsule as the library. Then publish the iTunes library. Now, all iTunes media player in your network will see the shared iTunes library.
TimeCapsule is a shared network storage. TimeCapsule is not an iTunes server.- Answered by Santhosh N from Ann Arbor
