No, you need Snow Leopard first. Lion is downloaded through the Mac App Store, which first appeared in Snow Leopard.
I think that to get Lion now, you need to contact Apple to obtain a redemption code, which then lets you download Lion.
Don't forget that, to run Lion, you need at least 2GB RAM (preferably more).- Answered by Rodney W from Niddrie

To download Lion or Mountain Lion you need to have at least Snow Leopard OS 10.6.8 installed.
There is no skipping... unless some Apple Genius at an Apple Store can install Lion or Mountain Lion from the store's network installer. But that might only be done under warranty if something happened to your hard drive and you needed it restored but had no backup of the OS.
Please note: as far as I know, Lion is not available for download since Mountain Lion is the current version of the OS. Unless Lion is available as legacy software from Apple by calling 1-800-MY-APPLE.
Lion used to also be available on a USB stick. I suppose if one had the USB stick you wouldn't need to have Snow Leopard to download Lion.- Answered by Ewald W from Vancouve

No, you can't skip over 10.6.x to get to Lion. Lion is only available through the Mac App Store. To get there you must get Snow Leopard and do upgrades to at least 10.6.6, where the Mac App Store is introduced and buy Lion from there.
