Yes it will I have the Otterbox Defender and it works.

Yes, I just got my Otterbox Defender case and the lightning to 30pin adapter and it works fine. The Otterbox Defender case leaves enough room for the adapter to fit.

This one will not fit (too wide), but the more expensive one with the 7" wire and smaller lightning connector works just fine.
