Can I set up the LaCie 2big as a Raid-1 and then direct Time Machine to it?

Yes. Use Apple Disk Utility to reformat and reconfigure the drive. Format as Mac OS Extended.
Then direct Time Machine to use the drive for backups.
You will only have 3TB of capacity when configured as RAID-1.
Inside are 2 3TB drives. In RAID-0, they are striped together to provide 6TB of fast storage.
In RAID-1, one 3TB drive is mirrored to the other 3TB drive.
